Abstract
In order to select one most divergent pair of parents for a mapping population, RFLP was used to detect polymorphisms among eight wheat varieties, including four accessions (“Alondra”, “Chinese Spring”, “Ningmai 3”, “Yangmai 3”) susceptible and four accessions (“Wangshuibai”, “Sumai 3”, “894037”, “895004”) resistant to scab. The results showed that the combinations of Wangshuibai/Alondra, Sumai 3/Alondra, 894037/Alondra had the highest polymorphisms and were suitable for constructing a mapping population. Now the recombinant inbred line (RIL)populations of these three crosses have advanced to F 5 generations with a total of 98 RFLP probes tested, the frequencies of the polymorphism were 30 6%, 28 6%,24 5% in above three mentioned combinations, respectively. When probe KSUF11 was hybridized to the genomic DNA digested with EcoRI, HindⅢ, BamHI, one specific band was observed in all four resistant varieties, but absent in susceptible cultivars, indicating the probe may be related to scab resistance. Two somaclonal variants were also identified with RFLP in this experiment.
